چکیده

n Thin-layer chromatography (TLC) is being used increasingly for the separation of the lipid classes (1, 2) prior to the analysis of their fatty acids by gas-liquid chromatography (GLC). Malins and Mangold (3) have described the separation of the methyl esters of fatty acids of menhaden oil from other lipid constituents and stated “the gas chromatogram of the total methyl ester fraction agreed in all details with a chromatogram obtained from a large scale preparation of these methyl esters.” This suggests that little if any autoxidation occurs during TLC. Identification of the spots is, of course, crucial to the eluting of the separate lipids. The use of iodine vapor is a favorite method (2, 3). It occurred to us that iodine may react chemically with unsaturated lipids, thus invalidating GLC studies of the eluted spots. The present study establishes this fact by showing a partial loss of polyunsaturated fatty acids after staining of the TLC plates with iodine vapor. * This research waa supported in part by PHS Research Grant AM

Preparative scale separation of neutral lipids and phospholipids by centrifugally accelerated thin-layer chromatography.

Mixtures of lipids and phospholipids were separated by centrifugally accelerated thin-layer chromatography on a preparative scale (300-500 mg lipid mixture per run). The isolated lipids and phospholipids were identified by 1H and 13C NMR spectroscopy and their fatty acid composition was determined by GLC and GLC-MS of their methyl esters.

Gradient-thickness thin-layer chromatography for the isolation and analysis of trace amounts of free fatty acids in large lipid samples.

A thin-layer chromatographic method for quantitative isolation of free fatty acids is described. This method appears to be more satisfactory than existing methods in offering the combination of advantages of specificity, simplicity, rapidity, reproducibility, accuracy, high sensitivity, and applicability as a preparative technique. The method involves chromatography on a thin-layer plate on whi...

Cerebrosides of human aorta: isolation, identification of the hexose, and fatty acid distribution.

Cerebrosides have been isolated from adult human aortic tissue. Each aorta was divided into portions classified as normal, fatty streaks, fibrous plaques, or complicated lesions. The cerebrosides were isolated by Florisil column chromatography, mild alkaline methanolysis, a second Florisil column, and preparative thin-layer chromatography. The concentration of cerebrosides was higher in fatty s...

Lipid composition in human breast milk from Granada (Spain): changes during lactation.

OBJECTIVE To determine possible differences of composition in the course of lactation, phospholipid (PL) classes (phosphatidylethanolamine, phosphatidylinositol, phosphatidylserine, phosphatidylcholine, and sphingomyelin) and fatty acid composition of PL and triacylglycerol (TGs) fractions of milk fat were analyzed in 66 samples from mothers from Granada (Spain) who gave birth to full-term infa...
